In vitro culture of grapevine meristem. I: 6-BA concentration in primary medium

Aimming at the development of a metodology for 'in vitro' grapevine meristem tip culture, the effects of four concentrations (1.0; 2.0; 5.0 and 10.0µM) of 6-BA (6 benzylaminopurine) cytokinin on growth and development of grapevine 'A Dona' isolated meristem tips in primary medium using basal medium of Nitsch & Nitsch were tested. Shoot tips were esterilized, meristems were then excised and after a brief period of pre-incubation were inoculated ira primary mediam. Viability, size, number of leaves and development speed of those meristem tips were studied. The concentrations of 2.0 and 5.0µM seemed to be the most appropriate to seedless A Dona cultivar.
