Sampling methods for starch determination in cassava hoots

SUMMARY

The following four sampling methods were compared in the determination of starch content in roots of individual cassava plants or in mixed roots from five plants chosen at random ; 1) a lengthwise sector 1/8 of the diameter was taken from every root in the sample ; 2) a sector was taken from every root in the sample as in 1) and then each sector was divided in three parts : bottom, middle, and top ; a 5-cm long cross section was taken from the middle of each of the three parts to compose the sample : 3) the roots in the sample were divided in two groups - thick and thin, and then ordered according to size ; from each group the median root was selected and a lengthwise sector 1/4 of the diameter was taken from the two roots and used for analysis ; 4) from each root selected as in 3), a 1/4 lengthwise sector was taken, and 5-cm long cross sections from the bottom, middle, and top were used to compose the sample.

Method 1) was used as a parameter since it included a proportional part of every root in the sample and thus eliminated the variation between roots and between parts of the same root.

Sampling method 3) gave the results closest to the parameter, following methods 2) and 4). Since samples obtained as in 3) were small and easy to prepare, this method is recommended as satisfactory for starch determination in cassava roots.

The results concerning number of plants in the samples were not conclusive. Preliminary evidence was obtained indicating that variation in starch content is greater between parts of the roots (top, middle, and bottom) than between the roots themselves.
