Indolbutiric acid concentrations and dark period on the in vitro rooting of blackberry (Rubus sp.), cv. ébano

The work was carried out aiming to test the influence of IBA and darkness on the in vitro rooting of blackberry cv. Ébano. Explants were used from the in vitro proliferation and they were subjected to two IBA concentrations (0.5 and 1.0mM) and three dark periods (2, 4 and 6 days). The explants were cultivated in a medium containing MS salt and vitamins (Murashige & Skoog, 1962) being the mineral salts reduced to half strength and added with myo-inositol (100 mg.L-1), sucrose (30 g.L-1) and agar (7.0 g.L-1). The pH was adjusted to 5.9. The flasks containning the explants after receiving the dark treatment were incubated in a growth room at 25±2ºC for 30 days. The root percentage, the number and root length as well as the callus formation in the shoot base were evaluated. After the plantlets being acclimatized it was evaluated the percentage of survival. No significantly differences for rooting were observed among the treatments. The root number was higher for media without IBA (5.5 roots/shoot). The longest roots were observed in the media without IBA and subjected to 2-4 days of darkness. The addition of IBA to the medium increased the intensity of callus formation. It was observed a higher percentage of shoots survival during the acclimatization (86-97%).
