RESUMO As causas da baixa tolerância à dessecação e baixa longevidade de sementes de café não foram ainda completamente elucidadas e o entendimento da sua complexa fisiologia é de grande interesse. Dentre várias alternativas, a perda da capacidade antioxidante nas sementes pode estar relacionada à rápida perda de qualidade durante o armazenamento. O objetivo neste estudo foi determinar a capacidade antioxidante total em café colhido em diferentes estádios de maturação, antes e após o armazenamento das sementes com diferentes teores de água, relacionando à qualidade fisiológica. Sementes nos estádios verde-cana ou cereja, recém colhidas ou armazenadas por nove meses em 10 °C com 40, 30, 20 e 12% de teor de água (bu), foram submetidas à avaliação da qualidade fisiológica, bioquímica e à determinação da capacidade antioxidante equivalente ao Trolox (TEAC). Observou-se que a germinação e a protrusão radicular de sementes de café nos estádios verde-cana e cereja não são afetadas pela secagem, mas sementes colhidas na maturidade fisiológica tem maior vigor com a redução da umidade. A qualidade das sementes de café é reduzida durante o armazenamento, sendo esta redução maior em sementes armazenadas com teores de água mais elevados. Sementes de café no estádio verde-cana tem maior capacidade antioxidante do que no estádio cereja quando recém colhidas, porém ocorre redução drástica desta capacidade durante o armazenamento, em ambos estádios de maturação. Em sementes de Coffea arabica L., os perfis isoenzimático do processo antioxidativo são pouco influenciados pelos estádios de maturação das sementes. A deterioração das sementes de café durante o armazenamento está relacionada à redução de sua capacidade antioxidante total, independentemente do estágio de maturação, sendo esta mais pronunciada no estágio verde-cana.

ABSTRACT The causes of the low desiccation tolerance and low longevity of coffee seeds have not yet been fully elucidated, and a full understanding of their complex physiology is of great interest. Among several alternatives, the loss of antioxidant capacity in seeds may be related to their rapid loss in quality during storage. The objective of this study was to determine the total antioxidant capacity of coffee harvested at different ripeness stages before and after the storage of seeds with different water contents and to relate antioxidant capacity to physiological quality. Seeds in the greenish-yellow or cherry stages, recently harvested or stored for nine months at 10 °C with 40, 30, 20 and 12% water content (wet basis - wb), were submitted to physiological and biochemical quality evaluations, and the Trolox equivalent antioxidant capacity (TEAC) was determined. The germination and root protrusion of coffee seeds from greenish-yellow and cherry fruits were not affected by drying, but seeds harvested at physiological maturity had greater vigor when the moisture content was lower. The quality of coffee seeds decreased during storage, and this decrease was greater in seeds stored with higher water contents. Coffee seeds in the greenish-yellow stage had a higher antioxidant capacity than those in the cherry stage when recently harvested, but there was a substantial reduction in this capacity during storage at both maturation stages. Coffee seed deterioration is related to a reduction in antioxidant capacity, and the isoenzymatic profiles of the antioxidant process are little affected by the seed maturation stage. The deterioration of coffee seeds during storage is related to a reduction in their total antioxidant capacity, regardless of their maturation stage, being more pronounced in the greenish-yellow stage.

RESUMO A criopreservação é uma técnica que pode potencialmente conservar o germoplasma de espécies do gênero Coffea por tempo indeterminado. O objetivo neste estudo foi avaliar os aspectos fisiológicos, bioquímicos e ultraestruturais de sementes criopreservadas de Coffea arabica L., cultivar Catuaí amarelo IAC 62, submetidas a diferentes protocolos de desidratação, pré-resfriamento, resfriamento, reaquecimento e uso de água catódica. De acordo com cada protocolo as sementes foram submetidas à secagem rápida ou lenta, até 17 ou 20% de umidade (base úmida), resfriadas por diferentes maneiras e, em seguida, foram imersas em nitrogênio líquido por 24 horas. Diferentes tempos de reaquecimento em banho-maria também foram utilizados. Análises fisiológicas, bioquímicas e ultra-estruturais foram realizadas nas sementes após as etapas de criopreservação. O teor de umidade a 17% bu é o fator chave para a criopreservação de Coffea arabica L., umidade em que as sementes apresentam melhor qualidade fisiológica e estruturas celulares mais preservadas. O pré-resfriamento das sementes de café antes da imersão em nitrogênio líquido é dispensado por não apresentar vantagens em relação à imersão direta. Os tempos de reaquecimento testados (2, 4 e 6 minutos) e o uso de água catódica não causam alterações na qualidade fisiológica e bioquímica, bem como nas estruturas celulares das sementes criopreservadas de Coffea arabica L. O padrão de estrutura celular observado em todas as sementes indica que o dano da criopreservação é menos drástico nas células dos embriões do que nos endospermas, sendo este último menos tolerante aos estresses de desidratação, pré-resfriamento e reaquecimento.

ABSTRACT Cryopreservation is a technique that may potentially conserve the germplasm of species of the Coffea genus for an indeterminate time. The aim of this study was to evaluate the physiological, biochemical and ultrastructural characteristics of cryopreserved seeds of Coffea arabica L., cultivar Catucaí amarelo IAC 62, which was subjected to different protocols regarding dehydration, precooling, cooling, rewarming and cathode water use. According to each protocol, the seeds were subjected to fast or slow drying to moisture contents of 17 or 20% (wet basis), cooled in different ways, and then immersed in liquid nitrogen for 24 hours. Different rewarming times in a water bath were also used. Physiological, biochemical and ultrastructural analyses were performed on the seeds after the cryopreservation steps. Moisture content at a 17% wb is the key factor for the cryopreservation of Coffea arabica L. seeds, which have better physiological quality and better preserved cell structures. Precooling of coffee seeds before immersion in liquid nitrogen does not provide advantages compared to direct immersion. The rewarming times tested (2, 4, and 6 minutes) and cathode water use did not cause changes in the physiological and biochemical quality or in the cell structures of Coffea arabica L. cryopreserved seeds. The pattern of cell structure observed in all seeds indicates that the damage from cryopreservation is less drastic in the cells of the embryos than in those of the endosperm, with the latter less tolerant to the stresses of dehydration, precooling, and rewarming.

RESUMO: As sementes recalcitrantes e intermediárias apresentam limitações com relação à conservação, devido a sua sensibilidade à dessecação e ao comportamento durante o armazenamento. Uma dificuldade no estabelecimento de metodologias para o armazenamento dessas sementes é devido à rápida deterioração, em que atuam processos que podem potencializar a produção de espécies reativas de oxigênio, causando danos oxidativos letais aos tecidos vegetais. Sementes ortodoxas, as quais apresentam maior tolerância à dessecação e ao armazenamento, também sofrem a ação dos processos deteriorativos, em graus variáveis com as espécies. Os danos causados por níveis nocivos de radicais livres podem ser amenizados pela ação de antioxidantes endógenos ou exógenos. Resultados de pesquisas recentes têm possibilitado novas tecnologias de proteção antioxidativa, sendo a proteção catódica uma técnica promissora e com resultados relevantes em espécies sensíveis à dessecação e em outros organismos vivos. Neste trabalho é apresentada uma revisão sobre a ação antioxidante da água catódica, produzida a partir da eletrólise de uma solução contendo cloreto de cálcio e cloreto de magnésio.

ABSTRACT: Recalcitrant and intermediate seeds have limitations regarding conservation because of their sensitivity to desiccation and response to storage. Establishing a method for seed storage is difficult because these seeds deteriorate rapidly, which can intensify production of reactive oxygen species that cause lethal oxidative damage to plant tissues. Orthodox seeds, which have greater tolerance to desiccation and storage, are also subject to deterioration processes, in degrees that vary according to the species. Damage caused by harmful levels of free radicals can be mitigated by the action of endogenous or exogenous antioxidants. Recent studies have led to new technologies for protection coming from antioxidants, one of which is cathodic protection. This is a technique with promising results in recalcitrant species, as well as in other living organisms. This paper reviews results regarding the antioxidant activity of cathodic water produced from electrolysis of a calcium chloride and magnesium chloride solution.

RESUMO: As enzimas têm papel fundamental na degradação de moléculas durante a germinação, desenvolvimento e deterioração das sementes. A endo-β-mananase é uma das principais responsáveis pela hidrólise das mananas no endosperma durante a germinação das sementes de café, atuando na degradação das paredes celulares e enfraquecimento do endosperma que circunda o embrião, permitindo a protrusão da radícula. Objetivou-se nesta pesquisa determinar a atividade da endo-beta-mananase nas estruturas das sementes de café, para avaliar a relação entre esta atividade e a qualidade de sementes submetidas a diferentes processamentos e métodos de secagem. Frutos de Coffea arabica L. no estádio de maturação cereja foram submetidos a três tipos de processamento: natural (sementes mantidas nos próprios frutos), despolpado (frutos descascados mecanicamente e sementes desmuciladas por fermentação em água) e desmucilado (frutos descascados e a mucilagem removida, ambos mecanicamente), como também a dois métodos de secagem: lenta à sombra (telado suspenso) e secagem rápida em secador mecânico a 35°C, até umidade de 11±1%. Após o processamento e secagem avaliou-se a qualidade fisiológica das sementes pelo teste de germinação e quantificou-se a atividade da enzima endo-β-mananase. Sementes de café submetidas ao processamento natural tem menor desempenho fisiológico, assim como maior deterioração e maior atividade da enzima endo-ß-mananase, principalmente após secagem mais rápida. A remoção da mucilagem durante o processamento despolpado e desmucilado de sementes de café reduz a atividade da enzima endo-ß-mananase e também a deterioração. A secagem rápida em secador afeta negativamente a qualidade das sementes. A enzima endo-ß-mananase é eficiente no estudo dos efeitos do processamento e secagem em sementes de café, podendo ser avaliada em sementes inteiras, endospermas ou embriões.

ABSTRACT: Enzymes play a fundamental role in degradation of molecules during seed germination, development, and deterioration. Endo-β-mannanase is one of the main enzymes responsible for hydrolysis of mannans in the endosperm during germination of coffee seeds through its action in hydrolytic degradation of cell walls and in weakening the structures of the endosperm that surround the embryo, allowing radicle emergence. The aim of this study was to determine the activity of the endo-β-mannanase enzyme in the structures of coffee seeds for the purpose of assessing the relationship between this activity and the physiological quality of the seeds under different processing and drying methods. Coffea arabica L. fruit in the cherry maturity stage was subjected to three different types of processing: natural (seeds maintained in the fruit itself), fully washed (fruit pulped mechanically and the seeds demucilaged by fermentation in water), and semi-washed or demucilaged (both fruit pulp and mucilage removed mechanically); and two methods of drying: slow drying (suspended screen) in the shade, and rapid drying in mechanical dryer at 35°C to a moisture content of 11±1%. After processing and drying, the physiological quality of the seeds was evaluated through the germination test, and endo-β-mannanase enzyme activity was quantified. Coffee seeds submitted to natural processing have lower physiological performance, as well as greater deterioration and greater activity of the endo-ß-mannanase enzyme. Removal of mucilage during fully washed and semi-washed processing of coffee seeds reduces the activity of the endo-ß-mannanase enzyme and lowers deterioration, especially after faster drying. The enzyme endo-ß-mannanase is efficient in studying of the effects of processing and drying on coffee seeds, and can be evaluated in whole seeds, endosperms or embryos.
