RESUMO Biosurfactantes, são emulsificantes naturais de hidrocarbonetos que diminuem a tensão superficial e têm alta capacidade de emulsionamento. Os biossurfactantes são produzidos pelo metabolismo de microrganismos e apresentam vantagens sobre o uso de surfactantes químicos, principalmente por sua baixa toxicidade e alta biodegradabilidade. A biorremediação é uma opção para a transformação de vários compostos contaminantes no solo e na água, sendo que os fungos são agentes transformadores eficazes, dada a sua capacidade de degradar uma ampla diversidade de substâncias orgânicas. Assim, considerando o potencial dos fungos na produção de biossurfactantes e a importância desses compostos para vários processos industriais, o objetivo deste trabalho foi verificar o efeito e as interações das concentrações de diferentes nutrientes na produção de biossurfactantes, utilizando três fungos coprináceos isolados do substrato pós-cultivo de cogumelos (spent mushroom substrate - SMS) resultante do processo de cultivo de cogumelos comestíveis. Os isolados fúngicos filamentosos codificados FS-4.1, FS-4.2 e FS-4.3 demonstraram potencial de produção de biossurfactantes, evidenciado pelo índice emulsificante no meio de fermentação. No entanto a produção biosurfactante foi máxima no isolado FS-4.1, produzindo um índice de emulsificante de 73,45%. Para otimizar o processo de produção biosurfactante FS-4.1 o óleo de milho, o glicerol e o extrato de levedura devem estar ausentes do meio de fermentação (para eliminar os efeitos negativos no processo) e aumentar as quantidades de glicose e óleo de soja.

ABSTRACT Biosurfactants are natural hydrocarbon surfactant compounds that decrease surface tension and have a high emulsifying capacity. Biosurfactants are produced through microbial metabolic processes and have several advantages over chemical surfactants, including low toxicity and high biodegradability. Bioremediation offers the possibility of eliminating or transforming various contaminant compounds present in soil and water. Fungi are effective transforming agents due to their ability to degrade a diverse variety of organic substances. Thus, considering the potential of fungi to produce biosurfactants and the importance of these compounds for various industrial processes, the objective of this work was to verify the effects and interactions of different concentrations of various nutrients on biosurfactant production in three coprinaceous fungi isolates. These isolates were obtained from spent mushroom substrate (SMS) produced during the edible mushroom growth process. All three fungal isolates, termed FS-4.1, FS-4.2, and FS-4.3, were potential sources for biosurfactant production, as evidenced by the emulsifier index of the fermentation medium. However, biosurfactant production was maximum in isolate FS-4.1, yielding an emulsifier index of 73.45%. To optimize the FS-4.1 biosurfactant production process, corn oil, glycerol, and yeast extract must be absent from the fermentation medium (to minimize negative effects on the process) and increase the amounts of glucose and soy oil.

RESUMO A produção de mudas de tomateiro é conduzida em substratos comerciais com propriedades adequadas para a boa formação da parte aérea e da raiz. O composto pós-cultivo de cogumelos (“Spent Mushroom Substrate” ou SMS) apresenta algumas vantagens em relação aos substratos comerciais para a qualidade das mudas de hortaliças, que podem ser proporcionados pela presença de uma rica microbiota, trazendo maior equilíbrio e maior competição com microrganismos patogênicos, além do controle biológico de patógenos e nematoides. É importante conhecer a microbiota presente nesse material e sua relação com a planta, para que essa associação possa ser feita da melhor maneira possível. Este trabalho teve como objetivo identificar a microbiota presente na rizosfera de mudas de tomate produzidas em SMS de cogumelos das espécies Agaricus subrufescens e Agaricus bisporus, acrescidos ou não de substrato comercial. A microbiota foi analisada por DGGE e amostras representativas de DNA foram sequenciadas para identificação das espécies. Entre os eucariotos, foram encontradas espécies de Chaetomium globosum, Arthrobotrys amerospora, predominantes no SMS de A. subrufescens, e Geastrum floriforme, Chrysosporium chiropterum, Pleurotus sp., e Trichosporon sp. no SMS de A. bisporus. Os procariontes apresentam maior diversidade, sugerindo um grande número de espécies ainda não classificadas ou cujas sequências ainda não estão disponíveis.

ABSTRACT The production of tomato seedlings is conducted on commercial substrates with adequate properties for the good formation of the aerial part and root. The Spent Mushroom Substrate, or SMS, presents advantages over commercial substrates regarding the quality of the vegetable seedlings, which may be provided by the presence of a rich microbiota, bringing higher balance and competition with pathogenic microorganisms, in addition to the biological control of pathogens and nematodes. It is important to know the microbiota present in this material and its relation to the plant, in order for this association to occur in the best manner possible. This work had the objective of identifying the microbiota present in the rhizosphere of tomato seedlings produced in SMS of Agaricus subrufescens and Agaricus bisporus mushrooms, added or not with commercial substrate. The microbiota was analyzed by DGGE and the representative samples were sequenced in order to identify the species. Among the eukaryotes, the Chaetomium globosum, Arthrobotrys amerospora species were predominant in the A. subrufescens SMS, and Geastrum floriforme, Chrysosporium chiropterum, Pleurotus sp., and Trichosporon sp. in the A. bisporus SMS. Prokaryotes are the most diverse group, suggesting a large number of species not yet classified or which sequences are not yet available.

Abstract Shiitake is an important edible and medicinal mushroom cultivated worldwide. Its cultivation involves a complex process of browning that precedes the primordia initiation. The present work through the evaluation of enzymes, lectin and β-glucan during the cultivation cycle of Lentinula edodes, the degradation of cellulose, hemicellulose and lignin, attempted to correlate these with mushroom yield. Strains UFLA-LE1, UFLA-LE2 and UFLA-LE6 consumed significantly more hemicelluloses than the other three strains with strain LE5 consuming significantly the most lignin of all six strains. Strains UFLA-LE4 and UFLA-LE6 were significantly the most productive. The laccase activity increased continuously until the end of the cultivation for all strains. Manganese peroxidase activities, except for UFLA-LE3, remained relatively stable. On the other hand, lignin peroxidase was the main lignin-modifying enzyme with much higher activity compared to laccase and manganese peroxidase. Tyrosinase activity was stable at an elevated level during the cultivation cycle, dramatically reaching the highest activity at the end of the cultivation period. The amounts of lectin and β-glucan varied greatly depending on the strain and the time of cultivation. Despite the many differences between strains in all evaluated parameters no direct association to the browning process was observed.

RESUMO A produção de Agaricus bisporus resulta em material residual significativo após seu cultivo. O substrato pós-cultivo do cogumelo (SMS), devido às suas propriedades físicas e conteúdo de nutrientes, tem um grande potencial para uso na agricultura. Nosso estudo avaliou a viabilidade do uso do SMS como substrato alternativo para a produção de mudas de tomate híbrido Paronset® e como fertilizante orgânico em sua produção. Para realizar o experimento de produção de frutos, foram utilizados três tipos de fertilizantes (SMS de A. bisporus, NPK e esterco bovino) com quatro repetições. As mudas produzidas no SMS apresentaram maiores índices de tempo e velocidade de germinação e se destacaram dos demais substratos na avaliação de qualidade de mudas. Na fase de produção de frutos de tomate, a média de produção das plantas originárias do SMS foi 20% maior que as mudas advindas do substrato comercial. As plantas cultivadas com o SMS apresentaram maior produção que as plantas cultivadas com esterco bovino incorporado ao solo. De acordo com nossos resultados, o SMS foi tão bom quanto o substrato comercial testado. Portanto, o SMS pode ser recomendado para o crescimento e nutrição da produção de mudas e denota potencial viabilidade para uso no ciclo de produção do tomate em suas diferentes fases.

ABSTRACT The production of Agaricus bisporus results in significant residual material after cultivation. Due to its physical properties and nutrient content Spent Mushroom Substrate (SMS) has great potential for use in agriculture. Our study evaluated the feasibility of using SMS as an alternative substrate for the production of Paronset® hybrid tomato seedlings and as an organic fertilizer in its production. To conduct the fruit production experiment, the plots consisted of three types of fertilizers (SMS of A. bisporus, NPK and bovine manure) with four replications. The seedlings produced in the SMS presented higher rates of germination time and speeds, and stood out from the other substrates in the evaluation of seedling quality. In the tomato fruit production phase, the average production of the plants originating from the SMS was 20% higher than the seedlings from the commercial substrate. The plants cultivated with the SMS presented higher production than the plants cultivated with bovine manure incorporated the soil. According to our results, The SMS was as good as the commercial substrates tested. Therefore, the SMS can be recommended for the growth and nutrition of seedling production and denotes potential viability for use in the tomato production cycle in its different phases.

RESUMO A compostagem é um processo de degradação natural da matéria orgânica realizado por microrganismos presentes no ambiente, levando a mineralização e humificação parcial das substâncias presentes na pilha, esse composto formado pode ser beneficamente aplicado ao solo como fertilizante orgânico na horticultura e agricultura. O número de estudos envolvendo inoculantes microbianos é crescente, os quais tem por objetivo a otimização de processos de compostagem. Contudo, o comportamento desses inoculantes e da microbiota ao longo do processo não tem sido caracterizado. Nesse contexto, este trabalho foi realizado com o objetivo de avaliar o efeito da utilização de um inóculo bacteriano que promova melhorias no processo de compostagem, bem como o de acompanhar a dinâmica populacional bacteriana ao longo de todo o processo através da técnica de High Resolution Melt (HRM). Para isso foram analisados quatro pilhas de compostagem inoculadas com Bacillus cereus, Bacillus megaterium, B. cereus + B. megaterium e o controle sem adição de inóculo. Foram realizadas análises químicas e moleculares (HRM) das amostras coletadas em diferentes períodos da compostagem (5º ao 110º dias). Os resultados mostraram que os inóculos bacterianos influenciaram no processo de compostagem com alteração na degradação de celulose, hemicelulose bem como alteração da temperatura e níveis de nitrogênio ao longo da compostagem. A utilização de um primer universal (rDNA 16S) permitiu acompanhar a sucessão bacteriana ao longo do processo, nos tratamentos. Contudo a construção de um primer específico é necessário para acompanhar de maneira mais precisa o inóculo durante o desenvolvimento da compostagem.

ABSTRACT Composting is the process of natural degradation of organic matter carried out by environmental microorganisms whose metabolic activities cause the mineralization and partial humification of substances in the pile. This compost can be beneficially applied to the soil as organic fertilizer in horticulture and agriculture. The number of studies involving microbial inoculants has been growing, and they aim to improve processes such as composting. However, the behavior of these inoculants and other microorganisms during the composting process have not yet been described. In this context, this work aimed to investigate the effects of using a microbial inoculum that can improve the composting process and to follow the bacterial population dynamics throughout the process using the high-resolution melt (HRM) technique. To do so, we analysed four compost piles inoculated with Bacillus cereus, Bacillus megaterium, B. cereus + B. megaterium and a control with no inoculum. The analyses were carried out using samples collected at different stages of the process (5th to 110th days). The results showed that the bacterial inocula influenced the process of composting, altering the breakdown of cellulose and hemicelluloses and causing alterations to the temperature and nitrogen levels throughout the composting process. The use of a universal primer (rDNA 16S) allowed to follow the microbial succession during the process. However, the design of a specific primer is necessary to follow the inoculum throughout the composting process with more accuracy.

Objetivou-se com este trabalho avaliar os efeitos inibitórios, "in vitro", de óleos essenciais dos condimentos, alecrim (Rosmarinus officinalis L.), cebola (Allium cepa L.), manjericão (Ocimum basilicum L.), menta (Mentha piperita L.) e orégano (Origanum vulgare L.), sobre o desenvolvimento de fungos. Os óleos foram extraídos pela técnica de arraste a vapor e testados nas concentrações de 500; 1000; 1500 e 2000 mg/mL-1. Como culturas de teste foram utilizados os fungos Fusarium sp.; Aspergillus ochraceus Wilhelm.; Aspergillus flavus Link e Aspergillus niger van Tieghem obtidos da micoteca do EcoCentro/EPAMIG em Lavras, MG. O óleo essencial do orégano inibiu o desenvolvimento dos fungos testados em todas as concentrações exceto o fungo A. niger que teve o seu desenvolvimento micelial inibido a partir da concentração de 1000 mg/mL-1,. Os óleos de alecrim, menta, cebola e manjericão tiveram um efeito pronunciado a partir da concentração de 1500 mg/mL-1.

The objective of this research was to evaluate "in vitro" effect of essential oils of the condiments, rosemary (Rosmarinus officinalis L. ), onion (Allium cepa L.), basil (Ocimum basilicum L.), mint (Mentha piperita L.) and oregano (Origanum vulgare L.) about fungi development. The essential oils were extracted by utilizing the vapor "dragging" technique and tested in the concentrations of 500; 1000; 1500 and 2000 mg/mL-1. The fungi Fusarium sp; Aspergillus ochraceus Wilhelm; Aspergillus flavus Link and Aspergillus niger van Tieghemwere obtained from the Fungal Culture Collection, EcoCentro/EPAMIG in Lavras, MG. The essential oil oregano inhibited completely the tested fungi development except the fungus A. niger that had its development inhibited starting from the concentrations of 1000 mg/mL-1. The rosemary, mint, onion and basil oils presented a pronounced effect starting from the concentration of 1500 mg/mL-1.
